Methodology
Methylmalonic Acid Urine (MMA) is analysed by chromatography-mass spectrometry (GC-MS) which is a powerful analytical technique used to separate, identify, and quantify complex mixtures of substances. It combines two methods: gas chromatography (GC) and mass spectrometry (MS). In gas chromatography, the sample is vaporized and injected into a heated column where different compounds separate based on their affinity for the column material and their boiling points. As each compound elutes from the column, it enters the mass spectrometer. In mass spectrometry, compounds are ionized and fragmented into charged particles. The resulting ions are then sorted based on their mass-to-charge ratio (m/z), producing a mass spectrum that serves as a fingerprint for each compound. By analyzing the retention times in GC and the mass spectra in MS, GC-MS can identify and quantify individual components within a mixture with high sensitivity and specificity.
